Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. Recognizes a protein of 33-37 kDa, identified as CD20 (Workshop V; Code CD20.12). B9E9 recognizes extracellular domain of CD20. The epitope is similar to or identical to that recognized by other CD20 antibodies including Leu-16 and B1. This MAb can be used for immunophenotyping of leukemia and malignant cells, B lymphocyte detection in peripheral blood, B cell localization in tissues and B lymphocyte purification by immunosorbent methods. CD20 is a non-Ig differentiation antigen of B-cells and its expression is restricted to normal and neoplastic B-cells, being absent from all other leukocytes and tissues. CD20 is expressed by pre B-cells and persists during all stages of B-cell maturation but is lost upon terminal differentiation into plasma cells. Reacts with five invariable CD3 chains designated CD3y, CD3d, CD3e, CD3f, and CD3n with molecular weight ranging from 16-28 kDa. CD3, also known as T3, is a member of the immunoglobulin superfamily that plays a role in antigen recognition, signal transduction, and T cell activation. It is found on all mature T-lymphocytes, NK, T-cells, and some thymocytes. It is expressed, typically at high levels, on peripheral T cells and majority of T cell neoplasms. Thymocytes express CD3 at different level on the cell surface in the course of differentiation and, in cortical thymus, CD3 is predominantly intracytoplasmic. Ten to forty percent of the patients with acquired immunodeficiency syndrome (AIDS) develop cytomegalovirus (CMV) infections. In some patients with AIDS, CMV is detected in the bronchoalveolar lavage fluid (BALF), urine, and other specimens, even when there are no symptoms of CMV disease. An indicator of active CMV infection is needed to facilitate the diagnosis of CMV disease in patients with AIDS or HIV infection. This antibody recognizes a protein of 30 kDa, identified as CD70. It is a cytokine that belongs to the tumor necrosis factor (TNF) ligand family. This cytokine is a ligand for TNFRSF27/CD27. It is a surface antigen on activated, but not on resting, T- and B-lymphocytes. It induces proliferation of co-stimulated T cells, enhances the generation of cytolytic T cells, and contributes to T cell activation. This cytokine is also reported to play a role in regulating B-cell activation, cytotoxic function of natural killer cells, and immunoglobulin synthesis. Recognizes an integral membrane glycoprotein of 150 kDa, identified as CD13 (also known as aminopeptidase-N). The antibody recognizes an extracellular epitope. The CD13 antigen is present on most cells of myeloid origin including granulocytes, monocytes, mast cells, and GM-progenitor cells. It is also expressed by the majority of AML, CML in myeloid blast crisis, and in a smaller fraction of lymphoid leukemias. CD13 is absent from normal lymphocytes, platelets and erythrocytes. CD19 is a transmembrane glycoprotein that contains two extracellular immunoglobulin-like domains. CD19 is present in both benign and malignant B-cells and is considered to be the most reliable surface marker of this lineage over a wide range of maturational stages. In normal lymphoid tissue, CD19 is observed in germinal centers, in mantle zone cells, and in scattered cells of the inter-follicular areas. Anti-CD19 exhibits an overall immunoreactivity pattern similar to those of the antibodies against CD20 and CD22. However, in contrast to CD20, expression of CD19 is continuous throughout B-cell development and through terminal differentiation of B-cells into plasma cells. Anti-CD19 positivity is seen in the vast majority of B-cell neoplasms commonly at a lower intensity than normal B-cell counterparts. 3, 3’ Diaminobenzidine (DAB) is a widely used chromogen for immunohistochemical staining. Betazoid DAB Buffer is one of two components used for DAB staining. This product is intended to be used in combination with Betazoid DAB Chromogen (BDB900C) available in the Betazoid DAB Chromogen Kit (BDB2004). The color of the chromogen buffer mixture may vary from colorless to pale brown. Staining properties are not affected by the color of the chromogen buffer mixture. In some cases, antibody titers may increase as much as 2-fold using this DAB system. Betazoid DAB can be used both manually and on automated stainers.
